چکیده

Table 1. The eflect of norvaline on citrulline synthesis Rat liver mitochondria (3.0 mg of protein/ml) were incubated in a medium containing lOOmM-KcI, 50mM-Tris/HCI, 1 mMEGTA, 1 mM-ATP, 20m~-succinate, 2pg of rotenone/ml, 5 mM-potassium phosphate, 16.6 mM-KHCO,, 10 mM-NH,CI and 3 m~-[~‘Clornithine (0.5 pCi/ml). After 4 min, mitochondria were separated from the suspending medium by silicone-oil centrifugation. Carbamoyl phosphate (CP) and ATP were determined in the neutralized HCIO, extracts. Parallel incubations with I “Clsucrose and ’H,O were carried out in order to determine the intramitochondrial ornithine concentration after separation of ornithine and citrulline by column chromatography.

[The role of adenine nucleotide translocator in the regulation of oxidative phosphorylation in heart mitochondria].

The regulatory role of adenine nucleotide translocase in oxidative phosphorylation was determined by titration of respiration of isolated rabbit heart mitochondria with carboxyatractyloside in the creatine phosphokinase ADP-regenerating system, which is not rate-limiting. It was found that the respiration rate is not controlled by adenine nucleotide translocase in states 3 and 4. Within the phy...

Modular kinetic analysis of the adenine nucleotide translocator-mediated effects of palmitoyl-CoA on the oxidative phosphorylation in isolated rat liver mitochondria.

To test whether long-chain fatty acyl-CoA esters link obesity with type 2 diabetes through inhibition of the mitochondrial adenine nucleotide translocator, we applied a system-biology approach, dual modular kinetic analysis, with mitochondrial membrane potential (Deltapsi) and the fraction of matrix ATP as intermediates. We found that 5 mumol/l palmitoyl-CoA inhibited adenine nucleotide translo...

Quantification of the contribution of various steps to the control of mitochondrial respiration.

Using principles developed by Kacser and Burns ((1973) in Rate Control of Biological Processes (Davies, D. Biochem. 42, 97-105), inhibitor titration studies were carried out in order to quantify the amount of control (control strength) exerted by different steps in oxidative phosphorylation on the ...

Mobilization of adenine nucleotide translocators as molecular bases of the biochemical threshold effect observed in mitochondrial diseases.

The existence of a biochemical threshold effect in the metabolic expression of oxidative phosphorylation deficiencies has considerable implications for the understanding of mitochondrial bioenergetics and the study of mitochondrial diseases. However, the molecular bases of this phenomenon remain unclear. We report here a new mechanism to explain this threshold effect, based on a reserve of enzy...

Effects of thyroid hormone on mitochondrial oxidative phosphorylation.

In order to locate sites of action of thyroid hormone on mitochondrial oxidative phosphorylation we have used an experimental application of control analysis as previously described [Groen, Wanders, Westerhoff, Van der Meer & Tager (1982) J. Biol. Chem. 257, 2754-2757]. Rat-liver mitochondria were isolated from hypothyroid rats or from hypothyroid rats 24 h after treatment with a single dose of...
